ABSTRACT:
Floating Drug Delivery Systems (FDDS) represent a significant improvement in gastroretentive technology, designed to prolong stomach residence duration and boost bioavailability for medicines with limited absorption windows. This review separates FDDS as effervescent systems, which employ gas production for buoyancy, and non-effervescent systems that rely on swellable polymers like HPMC and chitosan to maintain position. Manufacturing advancements, notably the combination of 3D printing with Hot-Melt Extrusion (HME), which enables the creation of customisable, pulsatile dosage forms. It also evaluates crucial formulation parameters, including polymer selection, excipient functioning, and physiological variables such as the fed or unfed state. Despite problems surrounding stomach emptying variability, FDDS offers a viable alternative for chronotherapeutic applications, eventually enabling increased therapeutic efficacy and adherence by patients.
